I have just checked two user accounts on each of three computers and they all show that Registry entry.
I also recall once setting up a new Windows 10 installation during which I deliberately looked at the User shell folders entries then used Screenshots for the first time and then looked again to witness that entry appearing in the Registry.
So I have no idea what is going on.
Are yours Local accounts or MS accounts? Have you ever changed any of the default user locations? None of my machine have that registry entry (or will create one when taking a screenshot) but all of them save to the Pictures\Screenshots folder. I've now checked back as far as v1511 on my System Two test machine. All my machines use Local accounts. Or it could just be that I've never changed any of the default location for my user files.

Maybe that registry entry is only used if the account is an MS account, or if user folders are changed from their default?. This thread suggests that deleting that registry entry will restore normal operation.

    ... delete the value named: {B7BEDE81-DF94-4682-A7D8-57A52620B86F}

The next screenshot taken should recreate the default folder in Pictures.

@NickosD could try deleting that value from the registry, restarting, then taking a WinKey+PrtScn screenshot. Export the key first, just in case.

Bree - I was wondering about the possible relevance of relocation but the OP's Registry Key includes what looks like a default location for that value. I use local accounts so I know it's not that.

Nickos - As long as you export the Registry key in .reg format first {right-click on the whole Key, Export}, I agree that it is worth deleting the entry we've been discussing to see what happens.

Bree - I have just checked a long-standing user account that has never had any of its user folders relocated and, like yours, it has no {B7BEDE81-DF94-4682-A7D8-57A52620B86F} entry. I am amazed.

Nickos - This reinforces the strength of Bree's suggestion about deleting that entry [after Exporting the Key as a backup]. Do you know why that entry was there? Did it appear when you started removing references to OneDrive?

Deleting the key did nothing. I still get the animation of taking the screenshot, but the registry key itself wont regenerate.

Cant access safe mode, it's broken at the current insider build.

I already did a repair of the installation, and after that I did a complete format. It might be a bug from the windows version. (But i had it before joining insiders too. Dunno)

Anyway, I am using greenshot for now.

Happy to hear that works. I presume you have it set to turn itself on at boot?

But the png file is not created in C:\Users\%UserName%\Pictures\Screenshots?

Have you restarted? Or at least signed out and back in? That is required for the registry changes to take effect.
Have you tried deleting the Screenshots folder? If deleted, does Winkey+PrtScn recreate it?

    Nickos,

Have you just clean reinstalled Windows 10? Not able to boot into Safe mode - it reboots when selecting the option

Have you ever had any reason to think there is anything wrong with your graphics driver?
Is it the latest available driver from the graphics card OEM and is it Windows 10 compatible for the current Windows 10 version?
Have you also done any available bios updates?

If you have not just done a clean install then, even if the graphics driver seems fine, reinstalling it will remove any concern that it is not operating correctly.

When did your screenshots problem start? After updating to a particular Windows version? After updating the graphics driver?

    Again,

I did a complete format already. Dont know why I just cant take screenshots with win + prnt but for know i use greenshot which does the same thing.

I did a restart after deleting reg key. It's strange behavior and for now I'll just wait for the next version of fast ring.

At the above safe boot issue I thought that I was just having an issue that's why I did a format on Friday.
