When a win 10 is on USB media

2) if that USB is valid for installing a new win 10 again, will I be able to upgrade to Win 10 Pro from it ?

Sorry for the late reply Cliff. I missed your post.
You will be able to use the ISO to install Windows with, or create a bootable USB with.
You can always use either to install Windows as many times as you like on any PC.
However, you will only be able to use either to upgrade to the free Windows 10 offer if it's for the same edition and version, and only until the July 29th 2016 deadline.

My Acer Aspire i5/5600u desktop and (2nd monitor) at home currently has a win 10 permanent activation. I am very interesting in learning a lot more about windows 10 and helping others as I learn. I would like to apply to be a Windows Insider and loading/checking some of the new Win10 builds. Please send me any help and additional info about being a Windows Insider to assist me.... and thanks. As of today I only have two more months to work outside and therefore more time for doing Win 10 and other things at home. Please consider.... V/r Cliff M.

Yes you can.
It may also be the case, if you skip entering the key, it may auto-activate as well.

My Bad - I should have added "to replace the outdated links to 'Latest' builds 10074 and 10162 in the top post" - which are respectively redirected to Technet evaluations for all versions of Enterprise, 10, 8, and 8.1, and the Insider Home page, which has little to do with Windows ISO preview file downloads.
