    Edit: Although the screenie shows MBR, each time I've changed it to GPT and Fat 32. It keeps jumping out of it, but I keep putting it back.
    So, you were successful in making the ISO from the ESD? If so, have you tried making a DVD instead of a USB stick?

    Wynona that error could for instance occur when you try to use an ISO which is at the moment mounted on your system as a virtual DVD drive.

    Could it be that you test the ISO's functionality first by mounting it, forget to eject / unmount it and try to use Rufus?


    EDIT:

    Checking Wynona's post again I noticed how wrong I was. That error message is not complaining about the ISO media being used but it tells that Rufus cannot use the USB media.

    Realizing that I started to play with Rufus and now I have the same issue! I have now tested three different USB Flash drives and I am getting the same error with each of them:

    The Rufus log file tells quite clearly that the problem is the access to the USB drive is denied. Have to do some research now to find out why.

    Wanting to test that the USB drives were not damaged causing Rufus not to get access to them, I downloaded and installed the Windows 7 USB Tool (Windows 7 USB/DVD Download Tool - Windows 7 Help Forums). With that tool I successfully created bootable USB install media using the same USB drives that Rufus can't use.

    Trying Rufus again, it shows the same error:

    I don't know what is causing this, why Rufus can't get access to the USB drive. The workaround, only working for those installing Windows on legacy BIOS systems is to use the above mentioned Windows 7 USB Tool (it does not create UEFI install media).

    Ah, forgot about UEFI - I drive an old-school BIOS rig. Another program that folks might try is UNetbootin for Windows - you have to manually format your USB stick to NTFS beforehand, but it works to make a bootable USB from the iso.

    Yeppers, but not in Build 10547!

    Sometime after I closed things down in here, I thought about dropping back to RTM Build 10240. So, this morning, I booted to it and just finished creating the bootable USB stick.

    So, at this point, it's looking seriously like Build 10547 could be the culprit; however, I haven't seen any other complaints about the inability to create the bootable media. Sigh . . .

    I see Kari's posted also, so I'll go read what he's found.

    Exactly, Kari. I thought it was talking about the ISO I created, but then I finally figured that one out. Still no joy after many other USB sticks.

    Could it just be an incompatibility between Rufus 2.4 and Build 10547? Reasoning is that each and every USB stick I tried all work without issue; I can add files, delete files, open files, close files, etc.

    Great minds and all that, Kari . . .

    I dropped back to my RTM Build 10240 and Rufus 2.4 worked without a hitch and I now have my bootable USB stick ready to clean install . . .

    Again, just for general information and to get everything I've tried in one place:

    1. Failure, as shown in my original screenshot.
    2. Tried second, third and fourth USB sticks: Failure
    3. First tries were on Desktop computer; subsequent tries were on Lappy 7: Failure
    4. Dropped back to Rufus 2.2: Failure

    A . n . d , drumroll please

    5. Booted into RTM Build 10240 and Rufus didn't hesitate: SUCCESS!




    Edit: Oh yeah, I also tried recreating Install.ESD to ISO: Failure
