Hello @abec99, and welcome to Ten Forums.

The registry keys for Windows Defender are protected by default.

You would need to change the owner of the registry keys to "Administrators", and set their permissions to allow "Administrators" "full control". Afterwards, you would be able to merge the .reg files.

However, doing that will leave the registry keys unprotected. This also will not help with the keys open by the system.

Hello Max, and welcome to Ten Forums.

Are you getting any error messages in PowerShell when trying to remove the exclusion for this path?

As test, see if you may be able to add the same exclusion, and try removing it afterwards.

It wouldn't hurt to do a full scan of your system using MalwareBytes just to make sure it's not infected.

    PS C:\WINDOWS\system32> Remove-MpPreference -ExclusionPath "C:" -Force
    PS C:\WINDOWS\system32> Add-MpPreference -ExclusionPath "C:" -Force
    PS C:\WINDOWS\system32> Remove-MpPreference -ExclusionPath "C:" -Force
    PS C:\WINDOWS\system32> Remove-MpPreference -ExclusionPath "C:" -Force
    PS C:\WINDOWS\system32> Remove-MpPreference -ExclusionPath "C" -Force
    PS C:\WINDOWS\system32> Add-MpPreference -ExclusionPath "F:" -Force
    PS C:\WINDOWS\system32>

No error messages. Also adding it, and removing it doesn't work. I also added F:\, which i could easily remove in the "windows defender exclusion window" by pressing remove. Also when i click on the remove Button of "C:" the first time it asks for Admin rights, but then nothing happens. When i click again on remove, nothing happens.

Downloaded the Software and clicked on Scan. Nothing was found.

    @MaxMicrosoftWin,

    This is an odd issue.

As a test, see if you may be able to remove the exclusion while in Safe Mode in case something running while in normal mode may be locking it.

In Safe Mode i can't open the Security Settings, it just loads a white Settings Page. I tried the Power Shell Commands in Safe Mode and restarted the PC but also didn't help.

Well, short of reinstalling Windows 10, you could try doing a repair install of Windows 10 to see if it may clear or at least allow you to remove it afterwards.
