Can anyone recommend a good software program that will move large folders (400G) from one drive to another drive without taking all day. Sometimes PC to USB but mostly USB to USB. I have been transferring all day (12 hours) and still have 3 hours to go transferring 816 GB.

No software going to speed up hardware transfer bandwidth speed.
You can plug a USB 2.0 device into a USB 3.0 port and it will always work, but it will only run at the speed of the USB 2.0 technology.
The built-in RoboCopy tool copies quicker than, for example, File explorer does because it multi-threads copying to make the most of the hardware's capabilities.
Example commands:
RoboCopy “F:\Whatever” “U:\Wherever” /S /R:10 /MT:32 /DCOPY:DAT /XA:SH
RoboCopy “F:\Whatever” “U:\Wherever” /S /R:10 /MT:32 /DCOPY:DAT /XA:SH /MIR
