I keep all important data on external HDD and periodically wish to synchronize this dats with another external HDD. Is this at all possible especially because drive letters themselves change depending on the PC/laptop that you attach them to and the order in which you attach them?

No problem! I like a program called FreeFileSync (FreeFileSync: Open Source File Synchronization & Backup Software). It can handle syncing regardless of the drive letter by specifying the volume name to get around changing drive letters. This program is free and has a few other really nice options that I like for syncing files.
However, if you do want to use something that insists on having the same drive letter, you should be able to work around this by assigning drive letters near the end of the alphabet. Other drives that you add will get letters such as d:. e:, f:, etc. assigned to them so they won't "step on" drives that have been assigned letters such as x:, y:, z:.
