If you select MBR in Rufus, I believe that you lose the option to boot in UEFI.
Actually I tried, I made a USB with mbr and was able to boot in uefi and make the disk gpt, the other way around if I make a USB just gpt and boot legacy it tells me invalid partition table, Anyways I’m gonna make my USB with Rufus gpt and make my bios configured to uefi only and test the difference.

It depends on what you set the Target System drop-down to. I know when making a Linux USB drive, when MBR was selected the only option available in the Target System drop-down menu was BIOS.
