Defragging a 2 Terabyte Drive
I have a 2 Terabyte drive of which only 357 GB is used, leaving 1.45 TB free.
With that much free space would it be beneficial to defrag my hard drive?

It is not about how much free space you have rather how fragmented the files you have are.
You can start the process and do the analysis portion of the defrag.

Win10, by default, will 'defrag' every week, in the background. No need to go out of your way.
I would grab defraggler to replace the default windows defragmenter. If your fragmentation is 20 percent or higher, defrag it.
Modern disc filing systems don't really need it, won't make any difference. Apart from that Windows 10 does it to a schedule. I set it to once per month. It does not get anything worse than a few percent with that. Weekly is more than enough.
I don't see any point in an alternative. Just more bloat doing very little.

actually these days defragging a disk of that size will take HOURS != and even then performance improvement will be minimal.
Why not just backup data, reformat the HDD and restore it -- depending on what data you have and its type this should work perfectly (backup data - don't clone or image the HDD).
In any case it's always good to have backup so this will serve as that too. If you have to image the HDD then use something like macrium free, and still re-format the HDD after taking the image. Mount the image file and then restore your data files with any appropriate program --even windows explorer !!!

My 2TB drive routinely defrags in 1-3 minutes.
- About 1TB in use
- I have it set to run daily just so it does not build up a great backlog of fragmentation to fix [I think it once took as long as 10 minutes when I had not defragged in a while]
- By sticking with Windows tools, I [automatically, by default] avoid moving fragments larger than 64MB [the threshold level used by MS is intended to avoid levels of defragging that would not yield any performance benefit].

Defragging an HDD can be done as often as you would like, no real harm in that.
So you have to ask yourself a few questions:
1) Do you have the time to wait?
2) It's mechanical in nature. More wear and tear.
3) Have you tried a program like Diskeeper that runs in real-time and hardly slows your computer?
Something to think about... for a boot drive!
