When you set DNS servers(primary, secondary) on your Router manually, you override those provided by your ISP, and for all networked PCs/devices.
When you set DNS servers in Windows it overrides those set in the Router so it is for the PC/device on which Windows is installed. Other devices will use those set in the Router.
If you set primary and secondary to different DNS services then the secondary(backup) will not get used.
That is my very simplistic understanding.
