Question
When creating rescue media on usb for macrium reflect, does that usb have to be left only for this purpose?
Once created and stored onto usb, can i move that rescue media file and store onto a hdd so i can use the usb stick for other files in the mean time?
No, you store the Rescue Media until you need it. That is the main point of making it and indeed checking that it works, and marking it so you know what it is for and how to Boot up from it.
They don't cost much, so no need to mess around.
No, once you have created a Macrium rescue usb you can continue to use it for data storage, create folders, copy files to it, etc.
In fact, as long as an existing usb is Fat32 format (as most are) then you can make it into a Macrium rescue usb without loosing any of the files already on the usb. Macrium does not reformat the usb as a matter of course, only if it's not Fat32. If it is already suitable Macrium just copies the boot files to it, no more than 1GB free space is needed for those.

I use the recovery media to create system images and of course restoring them
So really depending on a persons reflect use I don't even use reflect just the recovery media
Only quirk is I have to create a folder for the new system image go into but that's all.
I believe the system images created and verified with recovery media are more reliable than doing it in a active os.
That's my story and I'll stick to it
If the usb is large enough then you could store a system image on it too. A usb is perhaps not the most secure place to keep your only system image as they can fail more often than a usb HDD, but it is convenient if you need to do a restore while away from home. I have a 32GB rescue usb for my laptop that I take out and about with me, the highlighted files here were all added after making the rescue media. When making an image to a Fat32 usb Macrium automatically splits it into multiple 4GB files.
