    Quote for Tutorial above.
Under File system, select FAT32 (if able). If the file is larger than 4GB, you will only be able to select NTFS

What is the size of your ISO?

The size of the ISO file does not matter. The ISO file gets extracted to the USB flash drive, not copied to it.

Again, the size of the ISO file itself does not matter. The ISO file does not get copied to the flash drive. The contents of the ISO file get extracted to the flash drive.

Most UEFI computers require a FAT partition to boot from. RUFUS will make an "NTFS" USB flash drive that will boot in UEFI mode by creating a tiny FAT partition the computer boots from with files in it that then load the real boot files from the NTFS partition.

First....where did you get the ISO that you burned to the USB flash drive with Rufus?
You will have to disable secure boot and you do that in your UEFI (BIOS) settings. That's because you used Rufus to create the USB flash drive with.
As far as deleting C: drive - as long as your computer is booted from the USB flash drive and you are in Windows setup, you should delete all the partitions on your hard drive (or SSD) in your computer and then highlight the unallocated space and click next to let Windows setup create all the partitions on the hard drive it needs. Unless you have a separate partition created just to hold your data files, you would want to leave that one.
