Yes, the install thumb should be in FAT32. What I do is format the drive in Windows selecting the default option for the thump. I label it EMPTY so not to confuse with any others I may have plugged in. When complete then run Rufus to manage selecting the correct options for your machine of course..
Please use a camera or smart phone camera to take pictures of the BIOS menus > post images into the thread (If there are problems posting images then use one drive or drop box share links.
I'm curious when you deleted the partition did you by chance click the New option follow by apply ?

Someone else just asked about this problem. He was able to solve it by following a video. Here is the link to the thread. Read it all the way through.
We couldn't create a new partition or locate an existing one.

I am posting shots of the BIOS settings here. I do want to mention that I have tried again to do this install changing all settings back to how they normally are and creating the bootable ISO using Rufus with the MBR settings option and it still didn't work. I have not yet tried the manual settings using the CMD prompt options as that seems very complicated and my brain needed a rest before tackling that option which I will do today.
If you see anything in the shots of my BIOS settings that cause you to have any brilliant ideas for a solution please let me know...

I used this tutorial by Kari without creating a script and when I chose the newly created primary partition for Windows I got a message that read
"Windows cannot be installed on Disk 0 Partition 3 (see details)"
the details read
"Windows cannot be installed to this disk. The selected disk is of the GPT partition style."
Should I not be converting my disk to GPT as i suspected in the first place or is this message coming up because i changed my usb installation disk back to MBR in Rufus?

Getting the same message I see. Leads me to believe it's the thumb rather than anything. Not sure. Try burning a DVD.
A GPT scheme is only needed if it's a UEFI BIOS. You may check for sure, but I doubt if yours is.
I see you have it set for a GPT scheme, which may not apply.
Let me get a few things together. I'll be back.
Insert install media> Set BIOS to defaults and save > Hit the boot key for your PC > Pick the drive to start the install process > When the very first screen appears (OOBE screen) press SHIFT + F10 to open Command Prompt > Use this tutorial to clean the disk: Erase Disk using Diskpart Clean Command in Windows 10 Hardware Drivers Tutorials > Close out Command Prompt and continue with the install process. Let Windows manage it all. Pick and choose what features you want.
