Yes you need Defender on as well. Malwarebytes is not a full fledged anti virus product. I use the two of them together without issues. They target somewhat different items and complement one another nicely.
If you are using Malwarebytes along with an antivirus other than Defender, then no. I recommend downloading Avast if you want a free antivirus. Defender is not the best.
