Run CMD as admin and copy/paste:
    Code:
    reg delete "HKLM\Software\Policies\Microsoft\Windows Defender" /f

Try to open it manually: Run as administrator
    C:\Program Files\Windows Defender\MSASCuiL.exe

Try to reset Defender now in Services. Open Services in Admin mode and scroll down to Security Center
Right-click the Security Center service, and select restart.
    Restart your computer.

If you still have the same issue, then try this in cmd:
    1.Run the SFC scan
    Code:
    sfc /scannow
    2.
    Code:
    DISM.exe /Online /Cleanup-image /Restorehealth
Lastly, check if date and time are OK.

How do you get definition updates? WU?

    Manually, if not working:
Antimalware updates change log - Windows Defender Security Intelligence

    From cmd admin:

    1.
    Code:
    cd %ProgramFiles%\Windows Defender
    2. Shows definition that you have
    Code:
    MpCmdRun.exe -removedefinitions -dynamicsignatures
    3. Will generate an error if not working
    Code:
    MpCmdRun.exe -SignatureUpdate
    Latest at 2:40PM ET:

    Antimalware Client Version: 4.14.17639.18041
    Engine Version: 1.1.14800.3
    Antivirus definition: 1.267.1212.0
    Antispyware definition: 1.267.1212.0
Network Inspection System Engine Version: 1.1.14800.3
