Please type Powershell into Start Menu and start the Powershell as a administrator.

At the Powershell type below command and press Enter key.

    Code:
    Add-AppxPackage -register "C:\Program Files\WindowsApps\Microsoft.Windows.SecHealthUI_10.0.15063.0_neutral__cw5n1h2txyewy\appxmanifest.xml" -DisableDevelopmentMode
You may need to restart your computer.

The command @FreeBooter gave you was applicable to earlier versions of Windows 10. From the Creators Update, version 1703 the location has changed, the AppxManifest.xml is now to be found in:

    "C:\Windows\SystemApps\Microsoft.Windows.SecHealthUI_cw5n1h2txyewy\AppxManifest.xml"

The following command in Powershell (admin) should work.

    Code:
    Add-AppxPackage -register "C:\Windows\SystemApps\Microsoft.Windows.SecHealthUI_cw5n1h2txyewy\AppxManifest.xml" -DisableDevelopmentMode

I see @alpha45 has marked this thread Solved. Does that mean Defender now works properly for you?
